Been looking at some people's temple layout to do full runs plus Atziri every other run. Am I correct to assume that if the game allows me to place the key chamber where the red box is, and I lock down the 2 ends of the snakes, there would be no destruction?

Yeah, just make sure to only ever place it there, if you place it anywhere else, you will destabilise, also pre lock your lock to chain ends. Incase you crash or something while running temple, when you exit it still sometimes auto closes temple and if your not pre locked and have kill arch/atziri you will lose rooms. With this start you can only kill her every second run.
